Men
- Sky Masterson: (25-45) High baritone. Handsome, charming, self-assured gambler. Respected by other gamblers for the chances he takes.
- Nathan Detroit: (35-55) Baritone. Comical, lovable, engaged to Adelaide for 14 yrs. Arranges crap games and takes a cut of the action.
- Nicely-Nicely: any age, comic, a bit dense. Has the most notable number of the show; Sit Down, You're Rockin' the Boat.
- Benny Southstreet: Gambler
- Rusty Charlie: Gambler
- Harry, the Horse: Gambler
- Angie, the Ox: Gambler
- Big Jule: Gambler
- Arvide: Sarah's grandfather (55+) Has solo ballad, More I Cannot Wish You.
- Lt. Brannigan: (50+) police detective - always after Nathan to stop the crap games.
- ​Ensemble: gamblers, mission band and dancers.
Women
- Sarah: Soprano (20-40) Prim and proper Salvation Army officer, Has a hard time loosening up and adamant in her views.
- Adelaide: mezzo (35-45) Nathan's fiancee. Dancer in the Hot Box night club. Wants more than the dancer's life. Would like to settle down and have a "normal" life.
- General Cartwright: (45+) mezzo - Salvation Army general. Authoritative.
- Agatha and Martha: members of the Mission band
- Hot Box dolls: ( 20- 35) Must dance well and sing in the ensemble.
